Buffer The HeLa nuclear extracts have been collected in Lysis Buffer after a 30-minute incubation with TNF-α (20?ng/mL). The Lysis Buffer consists of 20 mM Hepes?pH 7.5, 350 mM NaCl, 20?% glycerol, 1?% Igepal-CA630, 1 mM MgCl2, 0.5 mM EDTA and 0.1 mM EGTA, 5 mM DTT and protease inhibitors. The protein content has been determined by a Bradford-based assay.

Handling Advice It ist recommended to aliquot the extracts into single-use fractions and then storing them at -80 °C. This eliminates repeated freeze/thaw cycles.
